阎小艳博士, 1980年出生,山西医科大学公共卫生学院教授、博士生导师。曾两次留学美国。山西省三晋英才、山西省优青、山西省优秀青年学术带头人、中国环境诱变剂学会青年委员、中国环境诱变剂学会环境应激与健康损害专委会委员、山西省青年科技协会常务理事、山西省环境诱变剂学会副秘书长。

获省级科技奖励4项,其中山西省科学技术奖自然科学类二等奖1项;山西省本科院校中青年教师基本功大赛二等奖,记二等功1次。参编出版著作两部。培养博士研究生1名,硕士研究生16名,其中2名荣获山西省优秀硕士论文奖,3名荣获校级优秀硕士毕业生称号。

主持国家基金3项,中国博士后基金1项;承担省级项目多项。在《Science of the total environment》、《Ecotoxicology and Environmental Safety》和《Archives of toxicology》等SCI收录的高影响力的期刊发表学术论文 56篇,其中第一或通讯作者23篇(SCI收录),其中13篇,212篇;非第一作者学术论文33(SCI收录),其中16篇,210篇。15篇代表性论文SCI他引检索为352次,被7种杂志广泛引用,其中13种,2区杂志2种。

曾前往美国、加拿大、波兰、伊朗等国参加国际学术会议并作会议报告8次;在中国毒理学会、中国环境诱变剂学会、中国地方病学会等举办的全国性学术会议中多次做学术报告,并引起同行的关注。

腹主动脉瘤是指腹主动脉局部或弥漫性的病理性扩张,是血管疾病中最凶险的一种类型,且发病率逐年攀升,造成了严重的社会和经济负担。目前主流治疗方法并发症多、患者经济负担大、再干预率高等,缺乏有效针对腹主动脉瘤的靶向治疗。基于上述难点,本项目在多项省部级基金支持下,针对腹主动脉瘤的细胞发病机制展开了一系列基础研究,取得如下创新性成果。

1. 建立腹主动脉瘤中平滑肌细胞(VSMCs)表型图谱,揭示新型腹主动脉瘤发病机制

VSMCs是血管中膜数量最多的细胞。早期实验发现VSMCs具有较高的可塑性。血管损伤刺激VSMCs转换为不同的状态或表型。近年来单细胞测序在血管衰老、动脉粥样硬化(AS)等疾病中发现了多种VSMCs表型,但当前尚无研究未对主动脉瘤中VSMCs表型进行分类以及鉴定。团队通过单细胞测序及细胞谱系示踪技术分析并鉴定出腹主动脉瘤中存在的主要6VSMCs表型,并明确各种VSMCs的细胞外基质、细胞黏附、炎症因子、蛋白酶等方面的功能。同时分析VSMCs各种表型的空间定位、转录因子、细胞通讯等性质。

2.创立小鼠VSMCs特异谱系示踪模型,追踪腹主动脉瘤中VSMCs命运

由于当前对VSMCs表型转换在腹主动脉瘤中的功能及命运所知甚少,团队在雄性Myh11-CreERT2+- tdTomato+/-小鼠上诱导腹主动脉瘤发生。作为首个追踪腹主动脉瘤中VSMCs分化命运的研究,团队发现由原VSMCs分化增殖而来的细胞失去了收缩型VSMCs的特点及标志物,转换为其他类型细胞,同时由动脉中膜迁移至内膜及外膜。这一发现突出了VSMCs表型转换在腹主动脉瘤发生中的重要作用。

3.构建AAV-miR-206-sponge,降低腹主动脉瘤扩张率及破裂风险,提供腹主动脉瘤治疗新靶点

基于VSMCs表型转换在腹主动脉瘤发生中的重要作用,团队筛选出腹主动脉瘤中与VSMCs表型转换相关的分子miR-206, 并针对性构建出AAV-miR-206-sponge以敲低miR-206。实现在小鼠腹主动脉瘤模型上降低瘤体破裂风险50%,降低瘤体最大直径约20%。团队实现了对腹主动脉瘤进行分子靶向治疗的可能性。

4. 通过外源性Tregs通过调节巨噬细胞极化稳定动脉粥样硬化斑块,减轻腹主动脉瘤危险因素

团队提前外源性Tregs细胞进行治疗减少动脉斑块面积,证明了Tregs细胞可以稳定颈动脉粥样硬化斑块,减轻动脉粥样硬化的发生及发展。

团队自进行相关基础研究以来,共计发表SCI论文16篇,最高影响因子7.5,被引次数:228次;授权专利6项。本项目有力地提高了我国血管外科在国际上的影响和地位。
